The role of different TRAF3 mutants in CD40 stimulation

Our lab studies the role of different TNF receptor associated factors (TRAFs) in lymphocyte activation. My work focuses on TRAF3. It is an adaptor protein that associates with CD40, BAFF-R to mediate various downstream singling events in activated B cells such as NF-κB1 & 2, JNK and ERK activation. TRAF3 function as a negative regulator of these receptors. Upon receptor stimulation TRAF3 is recruited to the receptor signaling complex, modified through ubiquitination and degraded by proteosomes. Therefore, mutations that are associated with TRAF3 will have a significant impact on these signaling pathways. A genome wide study of multiple myeloma patients performed by Dr. Bersagels’ lab identified several TRAF3 mutations, ranging from point mutations to large truncations. These mutations result in constitutive activation NF-κB2 pathway.

The aim of my work is to delineate how different TRAF3 mutants contributes to constitutive activation of NF-kB2 pathway and to further characterize which other signaling pathways are affected by different TRAF3 mutants.
